anti-Human CD3/CD28 Beads - 1 mL
T cells are activated in vivo by the first signal mediated by their T cell receptor (TCR) and the MHC molecule-antigen polypeptide complex on the surface of antigen-presenting cells (APCs), while the interaction (second signal) of the synergistic stimulation molecule CD28 on the surface of T cells with its ligand B7 (CD80/86) can enhance the activation and proliferation of T cells. T cells can be activated in vitro through the binding of anti-CD3 antibodies to CD3 molecules, and synergistic stimulation with anti-CD28 antibodies helps to enhance the activation effect. This product binds to the activating antibodies of CD3 and CD28 on magnetic beads to construct an artificial antigen-presenting magnetic bead with a 3D structure so as to achieve the effect of antigen presentation, thus achieving the purpose of activating T cells in vitro.
The magnetic beads of this product have a diameter of 1 μm, which can bind two antibodies to the membrane in equal proportions to form an artificial 3D antigen-presenting magnetic bead. The two antibodies selected for this product are humanized anti-CD3 and anti-CD28, both of which are GMP-grade products available from Novoprotein. Anti-CD3 is humanized from the finished drug OKT3 and targets CD3e; anti-CD28 can activate the co-stimulatory molecule CD28 on T cells.
1 mL of this product contains 4 × 109 magnetic beads and can be used to isolate 1000 mL of whole blood or 1 × 109 monocytes. The storage system is the phosphate (pH 7.4) buffer containing 0.1% HAS and 0.09% NaN3.
