An antibody specific for TNMD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNMD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for TNMD is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNMD bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:By searching an EST database for sequences similar to mouse Tem, followed by RACE of a fetus cDNA library, Shukunami et al. (2001) cloned TEM. The deduced 317-amino acid protein contains an N-terminal transmembrane domain and a putative antiangiogenic domain with 8 cysteines. TEM shares 96% amino acid identity with mouse Tem, and it shares 65% identity in a 65-amino acid C-terminal stretch with chondromodµLin I (CHM1). Northern blot analysis of several mouse tissues detected a 1.4-kb transcript expressed specifically in skeletal muscle. In situ hybridization revealed mouse Tem expression in dense connective tissue, such as epimysium and tendon. Stability:The stability of ELISA kit is determined by the loss rate of activity.
